總結(jié):
常用的單分子操縱技術(shù)— 微針、原子力顯微術(shù)、光鑷等存在作用力小且不易測(cè)量、實(shí)驗(yàn)環(huán)境
受限制、對(duì)生物樣品有損害等諸多不足。作為克服這些不足而出現(xiàn)的磁鑷技術(shù), 具有作用力大、應(yīng)用范圍廣、無(wú)損耗、穩(wěn)定等優(yōu)勢(shì), 很好得解決了這些問題。
磁鑷提供的作用力在0. 1—100pN 內(nèi)可調(diào),既可從分子外部進(jìn)行操作, 也可以深入細(xì)胞內(nèi)部, 還可對(duì)溶液體系中的鮮活樣品進(jìn)行操作。磁鑷應(yīng)用于生命科學(xué)研究已經(jīng)取得一系列成果, 如, 測(cè)定DNA 分子解旋過程拓?fù)洚悩?gòu)酶的作用[ 1] ; 測(cè)定鼠類巨噬細(xì)胞細(xì)胞質(zhì)的粘彈性[ 2] ; 分析核酸折疊動(dòng)力學(xué)[ 3] ; 測(cè)量牽引與粘合力[4] 等。
目前國(guó)內(nèi)對(duì)磁鑷的研究鮮有報(bào)道, 本文將介紹作者研制的磁鑷裝置以及利用該裝置進(jìn)行的實(shí)
驗(yàn)研究。
